Reduced loading times, improved ground game, submissions that aren't cribbed from Mario Party, counters that actually make sense instead of "YOU THREW A JAB NOW YOU GET SLAMMED", actual differences in fighting styles, characters that don't look like they're made of play do, a create-a-fighter that isn't awful, a career mode that isn't a tedious slog, the cage being something other than an irrelevant graphic, some sort of ability to get past Arona-style hold downs, commentary that isn't hideously one-sided and repetitive, BJJ that actually resembles BJJ, ragdoll physics that won't routinely turn Chuck Liddell's head through 360 or mean that Alves spends an entire fight with his elbow lodged in GSP's abdomen, music that isn't hideously grating, options that don't reset themselves occasionally, better sound effects, an audience that actually reacts to what's happening, tighter controls, faster gamespeed, more balanced fighters, removal of the various glitches and exploits and a clinch game that isn't hideously exploitable.
Of course, this being THQ, what we will get is a "karate" style (exactly the same as kickboxing but with faster jabs and weaker kicks), a roster update and maybe pictures of UFC fighters on the infinite loading screens.

I want an option that makes the opponent get up off his back and not butt scoot around or just lay there. This really pisses me off when im at a training camp and the opponent wont get up and I run out of time

Glad I'm not the only one who's noticed THQ's shitty track record when it comes to sequels (with the exception of Saint's Row).

-one or two new submissions (while taking away some, my money's on switching out the NS choke and mount triangle for yet another kimura variation and either the anaconda or d'arce choke)
-some gimmick dealing with either being rocked or the submission game (rotate the stick to clear the cobwebs and "defend intelligently").
-the addition of Kimbo and whoever wins TUF, but some other talent still missing (Bones, McLovin, Hardy, DHK, Sexyama, somebody's gonna be left out).
-fighter entrances and a TUF-based career mode, but the online will still be shit, quitting will still give both parties a DNF but no loss, the game will still have frame drops offline, and guys who get KOed will still do the ballerina spin while their mouthpieces fly.

Entrances for every fighter (They did it for the WWE, lets see some creativity here, Chuck bouncing with the music, Wand raising his hands for the fans, Sexyama pwning)
Pre-fight gear (Brand t-shirts, Gis, they had this kinda stuff in the PRIDE game and that was THQ)
Staredown controls (not amazingly important, but it would be cool to choose whether to touch gloves, snub them, whatever, just as a little extra for the personal touch)
Custom entrance music (give every fighter shitty studio music but leave the option to customise, that way we can edit it properly)
Revamped standup (Too limited, The K-1 World GP 2006 game on the PS2 had a pretty comprehensive cover of kickboxing for a button player, go from there, adding knees and the rest)
More submissions (Anaconda choke, Americana, D'Arce Choke, Heel hooks, Gogoplata, Omoplatas if only to hear Rogan say hes got it deep and then hes out again)
Rape choke (use it as leverage in your GNP, making it more effective)
Better physical damage (include doctor stoppages and leg kick TKOs, if Ive spent the last 5 minutes kicking a guy in the leg, he should be limping)
Improve the bottom game (Rubber guard was too powerful, apart from that being on the bottom seemed almost redundant, you should be able to tie them up to try and get a standup, rather than just holding their head)
Longer career mode (More automated created guys isnt a bad thing, just so long as Im not fighting at UFC 150: Machida vs Lowndes 7)
Freedom of weight at HW (235 wasnt good enough, there should be pros and cons for moving around the 60 pound limit, not everyone is Brock Lesnar)
PRIDE Legacy Pack (DLC perhaps, add the PRIDE ring, since they didnt use the cage, throw in the Saitama Super Arena as a location, fighters have the blue gloves, and even record some Japanese style crowd sounds, plus give a ton of fighters PRIDE-specific clothing, like Cro Cop having all checkered shorts, Nog having his old BTT spandex, Wands Chute Boxe black/whites, Rampage having all camo, Mark Coleman having the red shorts and kneepads)
Cage play (you should be able to push a guy against the cage and use dirty boxing, and take them down from the cage clinch, not to mention push them against the cage on the ground, and possibly use the cage to climb to your feet on the ground, make it some kind of struggle system)
